Oral Tolerance Therapy

Oral Tolerance Therapy
- Oral administration of protein antigens induces systemic immune tolerance.
- The two instruments hypothesized to introduced oral resistance are energy and dynamic invulnerable concealment.
- The acceptance of energy is best shown after inactivation or potentially cancellation of antigen-explicit T cells. By correlation, concealment is seen after various low-portion antigen taking care of.
- Trial information recommend that insusceptible concealment is accomplished through administrative cytokines, for example, TGF-b, IL-10, and IL-4. Immune system microorganisms creating these administrative cytokines may down regulate auto reactive T cells in an antigen vague way.
- This is useful in conditions like incendiary gut illness (IBD), in which pathogenic autoantigens have not been recognized.
- Oral organization of colonic antigens down regulates irritation in a rodent model of trinitrobenzene sulfonic corrosive (TNBS)- actuated colitis.
- Feeding of colonic extract proteins (CEP) induces suppressor T cells that mediate tolerance to tissue antigens. The conclusion was based on the up regulation of TGF-b in tolerized rats and the assumption that transfer of T cells from “tolerant rats” suppressed activities of endogenous T cells in recipient rats. However, it is difficult to determine how many endogenous T cells remained in irradiated recipients. In the absence of endogenous T cells, it is possible that donor T cells from “tolerant mice” failed to induce colitis because of pre-existing energy or immune deviation of cells toward a less pathogenic.
- For suppression to be demonstrated, they will need to show evidence that cytokines from donor cells downregulated effector cells important for colitis. Furthermore, if suppression did occur in recipient mice, the role of TGF-b will need to be studied in more detail.
- Overall, the adoptive transfer data are suggestive of a suppressive mechanism but will need more study if mechanistic conclusions are to be reached.
